ENDOTHELIAL FUNCTION AFTER EXPOSITION OF MAGNESIUM DEGRADATION PRODUCTS
in the HMT and MAN, the IC50 for both cases was not possible to calculate due to for the undiluted concentration the values were higher than 50%. However is possible to observe that around 100% of activity is reached around the third and the fourth dilution for all the samples Fig.2.
Fig.2. Serial dilutions of extract materials in HUVEC and SMC. Coating samples showed a lower release of Mg ions. After the third and fourth dilution the cell viability was not compromised.
3.2 Effect of Mg on endothelial cells under flow
Result from the experiments under flow conditions showed a more accurate effect of which can happen at the mo- ment of implantation. A slight increase in the pH of cell culture medium in presence of the materials was appreciated, however, this did not affect cell viability. Following 7-days in culture, the concentration of free Mg2+ in the medium was of 4.47 mM for c.p Mg, 3.21mM for HMT and 3.27mM for MAN. Corroboratively, the monolayer integrity of c.p. Mg was compromised, whereas the integrity of the monolayer of HUVECs exposed to the extracts from the coated material were intact.
Fig.3 Flow experiments under cells exposed to corrosion products of Mg. Monolayer of HUVECs were ob- served during 7 days to know the effect of Mg in the pH and integrity of the monolayer. Lower effects in comparison to what happened in static models was observed
