Shear stress modulates osteoblast cell and nucleus shape
Appendix A4
Figure A4. Effect of PFF on osteogenic differentiation of MC3T3-E1 osteoblasts. Prior to 1 h PFF, the medium was refreshed by a-MEM containing 2% FBS and antibiotics. PFF (amplitude: 1.0 Pa, peak shear stress rate: 6.5 Pa/s, frequency: 1 Hz) was generated for 1h using a pump and a parallel-plate flow chamber. Static control cultures were kept in a petri-dish under similar conditions as experimental cultures. Cells were post-incubated for up to 28 days. (A, B, C) The optical images show alkaline phosphatase (ALP) activity, collagen production, and mineralization after 21 or 28 days of cell culture. (D, E, F) Quantification of ALP activity, collagen production, and mineralization after 21 or 28 days of cell culture. CON, control; PFF, pulsating fluid flow. * p < 0.05.
