Regulation of bone cell mitochondrial structure and dynamics
RESULTS
Mitochondrial movement
Before PFF treatment, many mitochondria were distributed in the pre-osteoblast cytoplasm, particularly around the nucleus, and did not show obvious movement. During PFF treatment, mitochondria moved from left to upper right or right, corresponding to the flow direction (Fig. 3A). The number of mitochondria and their trajectory are shown in different colors. Within 2 min PFF treatment, mitochondria had changed their location in the cytoplasm, especially those located close to the cell membrane (Fig. 3B). Maximum track speed of mitochondria in cells treated by PFF yielded 14.9 μm/s, while minimum track speed was 0 μm/s (Fig. 3C). Most track speed values were in the interval 0, 2, and 4 μm/s. The 25 percentile was less than 0.75 μm/s, and the 75 percentile was less than 2.4 μm/s. Before PFF treatment, all track speed values were in the interval 0 and 2 μm/s.The 25 percentile was less than 0.47 μm/s, and the 75 percentile was less than 1.25 μm/s.
Determination of the track displacement of mitochondria revealed that the maximum and minimum displacements were 62.7 and 0 μm, respectively (Fig. 3D). Most of the track displacement values were in the interval 0, 10, and 20. The 25 percentile was less than 0.71 μm, and the 75 percentile was less than 4.8 μm. A schematic diagram of mitochondria in cells without and with PFF treatment illustrates that the changes in mitochondrial location can be seen clearly via simulation (Fig. 3E, F). Before PFF treatment, most of the track displacement values were in the interval 0 and 10. The 25 percentile was less than 0.53 μm, and the 75 percentile was less than 3.3 μm.
