Abstract:
Tyrosine-tags (G4Y) can be modified with SPOCQ for protein-protein conjugation, and is applied to introduce short-chain variable fragments (scFvs) and cytokines on antibodies in a modular fashion. Combined with sortase A ligation, trifunctional antibodies can be prepared by modifying both the light and heavy chains, generating a [2]:2:2 format, or by modifying asymmetrical knob- in-hole antibodies, generating a [2]:1:1 format.
